‘12A Railway Colony’ is a murder-mystery thriller centred around Karthik (Allari Naresh), a henchman working for a local politician. Karthik falls for his neighbor, Aaradhana (Kamakshi Bhaskarla), an aspiring sportsperson. The plot thickens when Karthik hides a mysterious parcel in Aaradhana's locked house and later discovers Aaradhana and her mother dead inside. The story then shifts into an investigative thriller as Karthik teams up with police officer Rana Pratap (Sai Kumar) to uncover the truth behind the twin murders, leading to several startling and unexpected twists related to the address and its history.
